Due to their toxicity, the increased distribution of microcystins (MCs) has become an important worldwide problem. MCs have
been recognized as inhibitors of protein phosphatase 2A (PP2A) through their binding to the PP2A catalytic subunit. However,
the exact mechanism of MC toxicity has not been elucidated, especially concerning the cellular response and its autoregulation.
To further dissect the role of PP2A in MC-induced toxicity, the present study was undertaken to determine the response of
PP2A in human amniotic epithelial (FL) cells treated with microcystin-LR (MCLR), one of the MC congeners. The results show
that a low-dose treatment of MCLR in FL cells for 6 h induced an increase in PP2A activity, and a high-dose treatment of MCLR
for 24 h decreased the activity of PP2A, as expected. The increased mRNA and protein levels of the PP2A C subunit may explain
the increased activity of PP2A. Furthermore, MCLR altered microtubule post-translational modifications through PP2A. These
results further clarify the underlying mechanism how MCLR affects PP2A and may be helpful for elucidating the complex toxicity

The development of ovarian follicular cells is controlled by multiple circulating and local hormones and factors, including
follicle-stimulating hormone (FSH) and epidermal growth factor (EGF). In this study, the stage-specific effect of EGF on FSH-induced
proliferation of granulosa cells was evaluated in the ovarian follicles of egg-laying chickens. Results showed that EGF and its receptor (EGFR) mRNAs displayed a high expression in granulosa cells from the prehierarchical follicles, including the large white follicle
(LWF) and small yellow follicle (SYF), and thereafter the expression decreased markedly to the stage of the largest preovulatory
follicle. SYF represents a turning point of EGF/EGFR mRNA expression during follicle selection. Subsequently the granulosa cells from SYF were cultured to reveal the mediation
of EGF in FSH action. Cell proliferation was remarkably increased by treatment with either EGF or FSH (0.1–100 ng/ml). This
result was confirmed by elevated proliferating cell nuclear antigen (PCNA) expression and decreased cell apoptosis. Furthermore,
EGF-induced cell proliferation was accompanied by increased mRNA expressions of EGFR, FSH receptor, and the cell cycle-regulating
genes (cyclins D1 and E1, cyclin-dependent kinases 2 and 6) as well as decreased expression of luteinizing hormone receptor
mRNA. However, the EGF or FSH-elicited effect was reversed by simultaneous treatment with an EGFR inhibitor AG1478. In conclusion,
EGF and EGFR expressions manifested stage-specific changes during follicular development and EGF mediated FSH-induced cell
proliferation and retarded cell differentiation in the prehierarchical follicles. These expressions thus stimulated follicular

Theaflavins (TFs) are the dimers of a couple of epimerized catechins, which are specially formed during black tea fermentation.
To explore the differences among four main TF derivatives (theaflavin (TF1), theaflavin-3-gallate (TF2A), theaflavin-3′-gallate (TF2B), and theaflavin-3,3′-digallate (TF3)) in scavenging reactive oxygen species (ROS) in vitro, their properties of inhibiting superoxide, singlet oxygen, hydrogen
peroxide, and the hydroxyl radical, and their effects on hydroxyl radical-induced DNA oxidative damage were systematically
analyzed in the present study. The results show that, compared with (−)-epigallocatechin gallate (EGCG), TF derivatives were
good antioxidants for scavenging ROS and preventing the hydroxyl radical-induced DNA damage in vitro. TF3 was the most positive in scavenging hydrogen peroxide and hydroxyl radical, and TF1 suppressed superoxide. Positive antioxidant capacities of TF2B on singlet oxygen, hydrogen peroxide, hydroxyl radical, and the hydroxyl radical-induced DNA damage in vitro were found.
The differences between the antioxidant capacities of four main TF derivatives in relation to their chemical structures were
also discussed. We suggest that these activity differences among TF derivatives would be beneficial to scavenge different

通过对黄山风景区浮溪河水域藻类植物的采集、室内显微镜检分类方法对该水域的淡水藻类资源进行调查研究。镜检过程中在所有样品中共记录到藻类植物112种(其中包括变种和变型),隶属5门42属。根据该水域藻类植物种群结构分析,知该水域中藻类植物群落属硅藻-蓝藻-绿藻-黄藻型。其中,硅藻在种类及数量上均占优势,有20属68种,占60.7%;蓝藻门,有11属21种,占18.7%;绿藻门,有9属16种,占14.3%;黄藻门1属4种,占3.6%;裸藻门,有2属3种,占2.7%。通过调查得知在沿河流自上而下采集的四个样品中,藻类的种类数以及植物体数量呈递增趋势。 相似文献

该文利用循环伏安法(CV)和线性扫描溶出伏安法(LSSV)研究了对苯二酚(HQ)和间苯二酚(RS)在聚噻吩/纳米二氧化钛修饰玻碳电极(PTh/NTiO2/GCE)上的电化学行为。该修饰电极作为两种苯二酚传感器表现出极好的灵敏度和选择性。在0.2mol/L柠檬酸-磷酸氢二钠缓冲溶液(pH 4.6),RS和HQ的氧化峰电位相距508 mV,且在PTh/NTiO2/GCE上的峰比在裸GCE上的高出6.5倍。在最佳条件下,PTh/NTiO2/GCE对HQ和RS在1.0×10-7~8.0×10-6范围内都有较好的线性关系,混合物中的检出限(S/N=3)分别为3.3×10-8 mol/L和3.7×10-8 mol/L。通过计算得出了一些动力学参数如电子转移数(n),质子转移数(m)。该法被用来同时测定废水中的RS和HQ结果满意。 相似文献
